Yes they will.
Not by much but I think there could be about an inch in it (dia).  7.50s are around 31in, 235/85s are nearly 32 BUT they all vary slightly between makes so its no exact science, especally if the old 7.50s are worn.

I wouldn't worry about the speedo calibration changing, asuming it was accurate in the first place.

my 7.50R16 michelins are the same height as my 235*85r16 goodyears

A fair bit wider but not much if any height difference.  Check your rims are wide enough for the 235's too........

As Jim-willy says, check the rims.

Your 750s will most likely be on 5.5" rims, the 235s I'm sure should be on wider rims like 6.5", though I can't lay my hands on that info at the moment.
Anyone know how wider Defender rims are?

What vehicle you got? For some you may have to alter the bump stops or the tyres will rub on the insides.
